Discover-sc WTA Kit V2 (N711) can use 1-1000 cells or 10 pg-10 ng total RNA as a template to obtain enough samples for sequence analysis through first-strand cDNA synthesis and amplification. It solves the technical problem that conventional mRNA-seq cannot be used for sequencing analysis of micro-samples such as single cells due to the low RNA content. N711 uses Oligo dTPrimer as the reverse transcription primer for cDNA synthesis, uses the Discover-sc Reverse Transcriptase Template-switching activity to add a linker sequence to the 3' end of the cDNA, and then uses the linker sequence for subsequent PCR amplification to obtain full-length cDNA amplification product. It is highlighted for the uniform transcript coverage and more gene detections, and therefore more suitable for the detection of low-frequency genes and low-concentration templates.
